If i manipulated a photo on photoshop, then flattened the image and saved it as a jpeg, is it impossible to tell that it's been edited in photoshop?

No its never impossible to tell. There are still tell tale signs no matter how good you are

There are people that can tell by close examination of the image, but, no, there's no flag in the file that says "she photoshopped-out a big zit on her nose" or anything like that.

There very often remain visual cues, and the EXIF data (within the file) often reflects that the file has been Photoshop modified to some degree.
